DDAB/PLA纳米颗粒佐剂浓度对免疫效果的影响

Abstract

目的以市售铝盐佐剂为对照,考察不同纳米颗粒佐剂浓度对rPA抗原免疫效果的影响,优化增强rPA抗原免疫效果的纳米颗粒佐剂浓度。方法采用间接ELISA、CCK8、流式细胞术、Luminex方法研究DDAB/PLA纳米颗粒佐剂浓度对免疫效果的影响。结果根据体外DC细胞、体内免疫后脾细胞体外孵育刺激后的活化及其细胞因子分泌水平结果,优化的纳米颗粒佐剂浓度为3 mg/ml;体内免疫后特异性抗体分泌水平结果显示,随纳米颗粒佐剂浓度增加,抗体分泌水平也提升;与铝盐佐剂相比,纳米颗粒佐剂对脾细胞的活化以及其细胞因子分泌的效果更佳。结论 3 mg/ml为纳米颗粒佐剂最佳佐剂浓度;纳米颗粒作为rPA疫苗佐剂可显著提高免疫效果。
