火炬松胚性悬浮细胞原生质体的分离和培养

Abstract以火炬松(PinustaedaL.)成熟合子胚的胚性悬浮细胞为材料分离原生质体,研究了酶液组成、渗透压稳定剂和悬浮细胞生长期对原生质体产量和原生质体活力的影响。结果表明:酶液组成为CelulaseRS1%+CelulaseR-102.5%+PectolyaseY-230.2%、渗透压稳定剂为甘露醇,在悬浮细胞处于对数生长期时,原生质体的产量和活力均最高。高活力的原生质体在DCR培养基上培养6d后完成第1次分裂,4周后形成胚性细胞团,8周后形成胚性胚柄细胞团(ESM:Embryonalsuspensormass),16周后胚性顶端分化出子叶原基。
